Philip Bolton attended Michigan State University, going on to the University of California, San Diego where he received his Ph.D. in chemical physics, working with Professor David R. Kearns on the structure of tRNA. He stayed on as Professor Kearns' postdoctoral fellow for two years, then worked for one year with Professor Thomas L. James at the University of California, San Francisco, studying the conformational fluctuations of RNA and DNA.

In 1979, Bolton joined the faculty of Wesleyan University and began working with two-dimensional nuclear magnetic resonance. His current research continues the study of the structures of quadruplex DNAs formed by aptamer, telomere and triplet DNAs. He is now looking at drug molecules which specifically bind to quadruplex DNAs, how potassium binding controls quadruplex structure and how loop length and composition affects quadruplex DNA structure. Bolton served as Chair of the Chemistry Department from 1995-1997. In addition to authoring over 100 articles for peer-reviewed scientific publications, he has participated in a number of site visits for the National Institutes of Health and the National Cancer Institute. He has also served on a number of study sections for NIH and the American Cancer Society.
